چکیده

IV-ethylmaleimide (NEM), an alkylating agent that covalently binds sulfhydryl groups, and dithiothreitol (DTT, Cleland's reagent), which reduces disulfide bridges to sulfhydryl groups, affected drug-induced contractions of rabbit aorta. NEM depressed aortic responses to potassium chloride (KC1), norepinephrine, serotonin, histamine, and angiotensin II. This action was attributed to an interaction of NEM with sulfhydryl groups vital to either the membrane function or the contractile apparatus. DTT had minimal effects on responses to KC1, norepinephrine, and serotonin; it potentiated aortic contractions produced by histamine and abolished responses to angiotensin II. Enhancement of the histamine response by DTT was not accompanied by an effect on the histamine-metabolizing enzymes or by an alteration in the KB and pA2 values for pyrilamine. The results suggest that a disulfide bridge plays a prominent role in both histamine and angiotensin receptor activity of rabbit aorta. With regard to the histamine receptor system, two possibilities exist: (1) reduction of a disulfide bridge at some vital point increases receptor activity or (2) the histamine receptor system, normally in a reduced state, is oxidized in the artificial environment of the tissue bath and needs to be reduced to regain full activity.

Evidence for the involvement of sulfhydryl oxidation in the regulation of fat cell hexose transport by insulin.

Previous studies have shown that the oxidants Cu(++), H(2)O(2), and diamide mimic the stimulatory effect of insulin on 3-O-methylglucose transport in isolated fat cells. The present experiments were designed to determine whether sulfhydryl oxidation plays a key role in the activation of the glucose transport system. It was found that reductants such as dithiothreitol inhibited 3-O-methylglucose...

Modification of cardiovascular responses to histamine by dithiothreitol.

1 Histamine produced dose-dependent contractile responses on both isolated perfused ear arteries and aortic strips of the rabbit. These responses were blocked by mepyramine and potentiated by both metiamide and dithiothreitol. 2 In the presence of maximum potentiation by metiamide, dithiothreitol still potentiated the contractile response to histamine of both preparations. 3 In the presence of ...

Oxidative-reductive modulation of guinea pig splenic cell guanylate cyclase activity.

The modulation of cyclic GMP metabolism by oxidativereductive-related processes was studied in intact guinea pig splenic cells and with respect to the activities of soluble and particulate guanylate cyclase. In intact cells the oxidant dehydroascorbic acid produced timeand concentration-dependent increases in cGMP steady state levels which were potentiated by the phosphodiesterase inhibitor 3-i...

Phenanthraquinone Affects Endothelial Nitric Oxide Synthase Activity through Modification of the Thiol Group: An Alternative Inhibition Mechanism

Phenanthraquinone (PQ) is a component of diesel exhaust particles which inhibits nitric oxide synthase (NOS) activity by shunting electrons away from the normal catalytic pathway of the enzyme and which can oxidize proximal protein sulfhydryls. In the present study, we examined the possibility that PQ may also modify critical thiol residues on endothelial NOS (eNOS), leading to a disruption of ...
